MyBioSource.com's Soluble Guanylate Cyclase alpha2 is a Rabbit Polyclonal antibody. This antibody has been shown to work in applications such as: Immunoprecipitation, and Western Blot. The Soluble Guanylate Cyclase alpha2 Antibody was generated using GUCY1A2 as the antigen and it reacts with Human, Mouse, and Rat.

Description

The cellular levels of Cyclic GMP (cGMP), a key messenger in several signal transduction pathways, are maintained by the orchestrated activities of two opposing enzymes: synthesizing gualylyl cyclases (GC) and hydrolyzing phosphodiesterases (PDEs). The synthesizing enzymes (GCs) are found in two forms: cytosolic (soluble) and membrane-bound (particulate). In last several years new members of particulate guanylate cyclase family have been identified and significant progress is made in understanding the molecular mechanisms that underlie the regulation of these family of enzymes. In response to G-protein couples receptor stimulation, the cGMP can be produced from GTP by either cytoplasmic, soluble guanylate cyclase (sGC) are heterodimers (á and polypeptide chains), that are stimulated by nitric oxide and carbon monoxide or by particulate membrane-bound guanylyl cyclases which are activated by a complex mechanism by natriuretic peptides. Particulate GC (PGCs) have 7 different isoforms, PGC-A through PGC-G and are expressed in most tissues in isoform specific manner (See Table 1). There is significant structural homology among various PGCs, there is a large N-terminal extracellular domain (ECD), a single TMD and a large intracellular domain with protein kinase activity (KLD), a C-terminal catalytic domain (CD) and in between is a dimmerization domain (DD). It has been shown that a significant number hippocampal astrocytes (67%) contained both soluble and particulate guanylate cyclases in the same cell (2). The Anti-sGCA2-selective antibodies were generated against conserved sequence near the carboxy terminal end of the protein (aa 692-708) that are unique to sGC-alpha2 subunit and is conserved in human with a single conservative substitution (Luecine to valine) in human protein. Soluble guanylate cyclase alpha1 is a 704 amino acid long protein. The sGCA2-selective antibodies are affinity purified against immobilized antigen based affinity chromatography which yielded epitope -specific antibodies. The sGCA2 antibodies label a 88-90kDa protein in soluble guanylate cyclase A2 Western blot positive control. Anti-sGCA2-selective antibodies are also available in affinity-purified form for confocal, WB and IHC analyses. MyBioSource will also conjugate antibodies with fluorescent probes upon request at extra charge. MyBioSource also provides antibodies to other family members of particulate GC (PGC-A, C, D, and G) and to adenylate cyclases (AC-1 through 9). MyBioSource, employs cyclic peptide methodology for generating high affinity antibodies (6). MyBioSource, also stock Western blot positive controls for most of these antibodies in ready-to-use buffer for easy identification of respective proteins. Limited quantities of antigens are also available for blocking studies. Please enquire for their availability before ordering
